CTR1: Bakır İthalatçısından Fazlası, Bir Sinyal Organizatörü

CTR1 as a transporter–organizer in copper signaling: molecular state, trafficking, and protein interactions

Meng‐Hsuan Wen, Tai‐Yen Chen

Kısa özet

Bakır Taşıyıcısı 1 (CTR1), sadece hücre içi bakır seviyelerini düzenlemek yerine, bakır akışını kendi durumuna, protein etkileşimlerine ve taşınmasına bağlayan bir "taşıyıcı-organizatör" olarak işlev görür.

Ana noktalar

  • CTR1'in işlevi, bakır ithalatının ötesine geçerek sinyal olaylarını organize etmeyi içerir.
  • Artan hücre içi bakır, CTR1'in oligomerik durumunu yeniden şekillendirerek bakır alımını akut olarak baskılar.

Copper transporter 1 (CTR1, encoded by SLC31A1 ) is best known as a high-affinity copper importer, yet several CTR1-dependent signaling outputs are not readily explained by bulk intracellular copper abundance alone. CTR1 has also been linked to growth-factor signaling, kinase regulation, redox control, trafficking, and copper-sensitive disease states. Here, we review recent evidence and propose a testable framework in which CTR1 can act as a transporter–organizer: a membrane protein that couples copper flux to transporter state, specific protein interactions, local copper handling, and trafficking history. Two observations provide the strongest experimental foundation. First, elevated intracellular copper rapidly remodels CTR1 oligomeric state before detectable internalization, with this remodeling associated with acute suppression of copper uptake. Second, CTR1 engages vascular endothelial growth factor receptor 2 (VEGFR2) through a redox-sensitive interaction that supports receptor signaling and co-internalization, while transport- and redox-defective CTR1 mutants functionally separate proximal VEGF signaling from canonical copper transport. Building on these observations, we discuss two testable hypotheses: transport-competent CTR1 may generate local copper-delivery windows that favor CTR1-proximal copper capture and exchange, and Rab-directed trafficking may tune the persistence and resetting of CTR1-associated signaling states. By separating established observations from testable models, this review outlines four variables—transport competence, local copper handling, specific protein interactions, and trafficking history—for dissecting CTR1-dependent outputs. More broadly, CTR1 provides a model for understanding how membrane transporters can coordinate metabolism, membrane traffic, and signal transduction.
